Expression of EZH2 in uveal melanomas patients and associations with prognosis

چکیده
PURPOSE To analyze the prognostic value and potential target for therapeutic intervention of enhancer of zeste homologue 2 (EZH2) in uveal melanomas (UM) patients. METHOD We analyzed EZH2 expression in 89 primary UM patients by immuno- histochemistry to observe the clinicopathological and prognostic value of EZH2. RESULTS The high levels of mitoses count and Ki67 labeling index had significant correlation with overexpression EZH2 (R = 0.408, P<0.0001; R = 0.72, P<0.0001). High level of EZH2 expression was significantly associated with increased risk of distant metastasis by the Cox proportional hazards regression model (multivariate hazard ratio: 2.12; log rank P = 0.037) and shorter UM-specific survival (multivariate hazard ratio: 3.92; log rank P = 0.036). CONCLUSION Our critical finding is that overexpression EZH2 in UM can be served as predictive marker and is associated with adverse clinical outcomes. Further observation of EZH2 as a potential therapeutic target in UM is necessary.

Effect of EZH2 Inhibition on Colorectal Cancer Cells: an In Vitro Study
Recently, the epigenetic modifications have been recognized as a regulator of gene expression in various cancers. EZH2 gene is one the most important component of the PRC2 complex. Overexpression of EZH2 was identified in multiple cancers that considered more attractive the EZH2 role as an oncogene. Some studies report that EZH2 contributes to various aspects of colorectal cancer (CRC). However...
متن کاملDysregulated Expression of Long Non-Coding RNA MINCR and EZH2 in Colorectal Cancer
Background: As critical regulators, lncRNAs have attracted attention from researchers for diagnostic, prognostic, and therapeutic purposes in human carcinogenesis via interfering with mRNAs such as EZH2. Nevertheless, the potent roles and molecular mechanisms of these RNAs in CRC are not clearly known. Methods: In this study, the tissue expressions of lncRNA MINCR and EZH2 mRNA between colorect...

PURPOSE To evaluate the role of nm23 gene expression in the development of metastases of human uveal melanomas in an animal model. METHODS Seven human uveal melanoma cell lines and two murine skin melanoma cell lines were subjected to Northern blot analysis for the detection of nm23-H1 mRNA and to immuno-histochemistry to detect nm23 antigen. Each tumor cell line was transplanted intracameral...
متن کاملOcular melanoma: an overview of the current status.
Ocular melanoma is the second most common type of melanoma after cutaneous and the most common primary intraocular malignant tumor in adults. Large majority of ocular melanomas originate from uvea, while conjunctival melanomas are far less frequent. Incidence of uveal melanoma has remained stable over last three decades. Diagnosis is in most cases established by clinical examination with great ...
متن کاملHeat shock protein expression in the eye and in uveal melanoma.
PURPOSE Expression of heat shock proteins (HSPs) is of prognostic significance in several tumor types, whereas HSPs may also have clinical use as stimulators in tumor vaccination. HSP expression levels were determined in normal eyes and in uveal melanoma and tested whether HSPs expression was associated with prognostic parameters in the uveal melanoma. METHODS Expression of HSP27, HSP70, HSP9...
